Buffer solution
Dissolve 10 g of sodium dodecyl sulfate and 20 mL of glacial acetic acid in 1.2 liters of water, and mix.
Mobile phase
Prepare a filtered and degassed mixture of Buffer solution and acetonitrile (33:17). Make adjustments if necessary.
Standard preparation
Dissolve an accurately weighed quantity of
USP Hydromorphone Hydrochloride RS in water, and dilute quantitatively, and stepwise if necessary, with water to obtain a solution having a known concentration of about 40 µg per mL.
Assay preparation
Weigh and finely powder not fewer than 20 Tablets. Transfer a portion of the powder, equivalent to about 4 mg of hydromorphone hydrochloride, to a 100-mL volumetric flask, dissolve in and dilute with water to volume, sonicate if necessary, and mix. Filter a portion of the solution using a glass fiber filter, and discard the first 5 mL.

Procedure: the tailing factor for the hydromorphone hydrochloride peak is not more than 2.0; and the relative standard deviation for replicate injections is not more than 3.0%.
Procedure
Separately inject equal volumes (about 100 µL) of the
Standard preparation and the
Assay preparation into the chromatograph, record the chromatograms, and measure the responses for the major peaks. Calculate the quantity, in mg, of hydromorphone hydrochloride (C
17H
19NO
3·HCl) in the portion of Tablets taken by the formula:
0.1C(rU / rS),
in which
C is the concentration, in µg per mL, of
USP Hydromorphone Hydrochloride RS in the
Standard preparation; and
rU and
rS are the peak responses obtained from the
Assay preparation and the
Standard preparation, respectively.
